K3 edta when added to PBS at pH 7.5 at 1.8mg/ml (blood tubes) leaves you with a pH of 7.5, when using K2 edta your final pH is 6.8. No idea how it effects other buffer systems.
 
Dipotassium EDTA and tripotassium EDTA; that is the only difference. However, when you refer to PCR, I believe that you are talking about the low concentration present in the enzyme (0.1mM). It is simply added to prevent the poisoning of magnesium-dependent enzymes with irrelevant metal ions like Pb2+. If such metal ions are present, it will bind to your enzyme and not the magnesium ions. At such minuscule concentrations, K2 and K3 have no significant difference.
 
Automated instruments react in a different way, their MCV is not influenced by K*-EDTA concentrations up to ten times normal, while K*-EDTA, at high concentrations, results in a slight increase in MCV, as measured with three of the instruments.
